Conference: ASMS 1998 Session Code: WPD Slot: 175 Title: Automated Protein Identification by MALDI-MS Authors: *Clauser, Karl; Tang, Chris; Rosenberg, Stan; Baker, Peter; Hsieh, Frank; Institutes: Abstract: Delayed-extraction and reflectors have improved the mass accuracy obtainable on MALDI-TOF instruments almost to the point where elemental composition can be routinely determined for each component in a peptide mixture. Hence, when peptides are derived from digestion of 1-3 proteins in a mixture and the sequence of those proteins exist in a genome database, peptide mass fingerprinting allows high-confidence identification of the proteins. We have developed an integrated system for automating the portions of this process following protein isolation. The following components will be described: digestion, de-salting, MALDI target spotting, spectral acquisition, spectral processing, and database searching.
